Plasma-SeqSensei™ Colorectal Cancer RUO Kit

Das Plasma-SeqSensei™ (PSS) CRC RUO (research use only) Kit erlaubt die hochsensitive und spezifische Erkennung von Mutationen zirkulierender Tumor-DNA (ctDNA) im Plasma von Patienten mit kolorektalem Karzinom.

Das Kit basiert auf Next-Generation-Sequenzierungstechnologie und deckt Schlüsselmutationen der MAPK-Signalgene KRAS, NRAS und BRAF sowie die PIK3CA-Gene ab.  Diese Gene tragen signifikant zur Entstehung von Darmkrebs bei und sind wichtige Biomarker für Prognose, Therapiewahl und Monitoring von Rezidiven und Therapieansprechen. [1]

KRAS ist in ca. 40% aller Fälle von kolorektalem Karzinomen mutiert (in Exon 2, Codons 12 (70-80%) und 13 (15-20%)). Die verbleibenden Mutationen sind meist in Exon 3 Codons 59-61 lokalisiert und in Exon 4 (Codons 117 und 146)). [2]

Mutationen in NRAS werden in 3-5% aller Darmkrebsfälle beobachtet (in Exon 3 Codon 61 (60%) und in Exon 2 Codons 12 und 13). NRAS-Mutationen und solche in KRAS schließen sich normalerweise gegenseitig aus. [3]

BRAF-Mutationen (gewöhnlich V600E) liegen in 8-12% der Patienten mit metastasiertem kolorektalen Karzinom vor, fast nie liegen gleichzeitig RAS-Mutationen vor. [4-5]

Die Häufigkeit von PIK3CA-Mutationen bei Darmkrebs liegt bei 7-32%, die Mutationshotspots liegen dabei in Exons 9 und 20. [6]

Für die Konfiguration des Plasma-SeqSensei™ CRC RUO (research use only) Kits wurden basierend auf den angegebenen Mutationsfrequenzen in den Datenbanken COSMIC (Catalogue of Somatic Mutation in Cancer) und cBioPortal for Cancer Genomics diese vier Gene als wichtige Schlüssel-Onkogene ausgewählt.
